Greetings! I am wondering if anyone has compared the efficiency of
different extraction procedures on recovering polyunsaturated fatty acids.
I have tried several common methods (Folch, Dole, etc.) and recovered nearly
100% of palmitate or oleate, but found such fatty acids as 20:5 and 22:6 to
have poor recovery (70% at best).
I became aware of this problem when I tried to measure acid-soluble products
in some in vitro incubations with a variety of different radiolabeled long
chain fatty acids. What I found is that the acid precipitation of the fatty
acid/protein complex was not the same between fatty acids. So now I have
media with radiolabeled fatty acids, which are contaminating my acid-soluble
products measurements. Any thoughts on how to separate the two or
effectively extract PUFA would be greatly appreciated.
