You have globally two solutions :
1- saponify the lipid extract (as described in
cyberlipid.org)in the presence of antioxidants and
separate the tocopherols by reversed-phase
chromatography.

2- separate the crude extract, even directly the oil,
by normal phase chromatography

Yes there are several synthetic inhibitors of fatty acid desaturases, you will find a lot of references in screening PuMed with Fatty, desaturase and inhibitor...
