Dear Mr Herrera
Your question about the volume of the lower phase has
no practical importance with the lipid extraction
process. You must remove the upper or the lower phase
completely and accurately. It's not complicated and
the variation of the result is very low when the
volume is about 4-5 ml. With Pasteur pipette you may
remove or collect the phases very accurately. You can
also use phase separation filter paper for larger
volume, but some contaminants (hydrocarbons) are
eluted in the extract.
You may replace chloroform by dichloromethane, the
results will be similar.
